摘要
目的探讨核转录因子-κB(NF-κB)、诱导型一氧化氮合酶(iNOS)蛋白在结肠癌组织中的表达、相互关系及其与临床特征之间的关系。方法采用免疫组化PowerVisionTM两步法检测76例结肠癌组织中NF-κB、iNOS的表达情况。结果NF-κB、iNOS结肠癌组织中的表达率分别为57.89%(44/76),69.74%(53/76)。NF-κB、iNOS在结肠癌中的表达与肿瘤大小、分期及淋巴结转移有关,三者均与性别、年龄、肿瘤类型无关。在结肠癌中,NF-κB、iNOS表达具有相关性。NF-κB、iNOS阳性组微血管密度(MVD)高于阴性组MVD,两者比较具有统计学意义(P<0.05)。结论在结肠癌组织中,高表达的NF-κB与肿瘤的发生发展有关,而NF-κB调节下高表达的iNOS亦与肿瘤血管生成乃至转移密切相关。
Objective To explore the expressions of nuclear transcription factor KB ( NF -κB) and inducible nitric oxide synthase (iNOS) in colon cancer and identify the correlation of the expressions of NF -κB and iNOS with the clinical features. Methods The expressions of NF -κB, iNOS and its correlation with the clinical features were determined using immunohistochemistry ( Power VisionTM two step methods) in 76 cases of human colon cancer. Results The expressions of NF-κB and iNOS in colon cancer tissue were 57.89% (44/76) and 69.74% (53/76) respectively. Overexpression of NF-κB and iNOS in colon cancer was related to lymph node metastasis, tumor staging and tumor size ( P 〈0. 05 ), but not correlated to age, sex or histological classification. The expression of NF-κB in colon cancer tissue was positively correlated with iNOS expression (P 〈0. 05 ). The microvessel density (MVD) in NF-κB and iNOS positive groups was significantly higher than that in negative groups ( P 〈0. 05 ). Conclusion The overexpression of NF -κB plays an important role in the development of colon cancer, while the expression of iNOS, upregulated by NF-κB, is also an important role in the angiogenesis and development of colon cancer.
