摘要
本研究利用反转录-聚合酶链式反应(RT-PCR)检测了ndrg2 mRNA在人胎脑组织中表达的时空特点,为进一步研究其功能提供实验依据。收集正常引产16~28周人胎脑组织标本,进行总RNA提取,应用RT-PCR方法半定量分析ndrg2基因在上述人胎脑组织中的表达水平和变化规律。RT-PCR结果显示:ndrg2 mRNA在正常人胎脑16、20、24和28周的各脑功能区均有表达,但随着发育,其表达水平有差别。在小脑、延髓和海马等部位,ndrg2 mRNA的表达水平在16周胎脑中较高。而在额叶和顶叶等部位,ndrg2 mRNA的表达水平在28周胎脑中达到高峰。以上结果表明,在人胚脑发育过程中ndrg2 mRNA从16~28周均有表达,但在不同脑后其表达存在一定差异,提示ndrg2基因对中枢神经系统的发育和成熟可能有重要影响。
In the present study, reverse transcriptase polymerase chain reaction ( RT-PCR) was used to detect the temporal and spatial expression of ndrg2 mRNA during human embryonic brain development, and to provide some clue for further study the function of ndrg2. Human embryonic brain tissues of induce-production from the 16th to the 28th week were collected. Total RNAs were extracted from human embryonic brain tissues. RT-PCR was used to quantitate the relative abundance of ndrg2 mRNA expression and analyze the changes. RTPCR results showed that ndrg2 mRNA was found to be extensively expressed in the normal embryonic brain tissues at the 16th to 28th week. With the development of human embryonic brain tissues, the levels of ndrg2 mRNA expression in each embryonic brain functional areas had different, ndrg2 mRNA expression in cerebellum, medulla oblongata and hlppocampus of 16th week embryonic brain increased significantly. After 28th week, ndrg2 mRNA expression in frontal lobe and parietal lobe arrived at peak. These results indicated that ndrg2 mRNA was expressed in the normal human embryonic brain from the 16th to 28th week, but the levels of expression in different areas varied, suggesting that ndrg2 might be related with the development and the mature of human central nervous system.
