蜂胶及自制蜂胶奥硝唑合剂对人牙龈成纤维细胞的毒性作用 被引量:2

摘要 目的:了解蜂胶及自制蜂胶奥硝唑合剂对人牙龈成纤维细胞的毒性作用,为蜂胶及合剂的临床推广应用提供理论依据。方法:人牙龈成纤维细胞在不同浓度的蜂胶、奥硝唑和自制蜂胶奥硝唑合剂中体外培养24h,用MTT法测定细胞相对增殖率(RGR);选择细胞增殖率约为50%的药物浓度组,重新培养细胞24h,更换成不含药物的培养液连续培养7d,每天用MTT法测定吸光度值,了解细胞增殖回复情况。结果:0.5g/L蜂胶组的RGR达到93.9%,与蜂胶其他2组间的差别有统计学意义(P<0.05);奥硝唑溶液加入蜂胶后,对应浓度组比较,两合剂组的平均吸光度值均大于奥硝唑组,其中当奥硝唑浓度为0.4g/L和0.8g/L时,差别有统计学意义(P<0.05);细胞增殖率约为50%的药物浓度组更换新鲜培养液后,细胞数均持续增加,到第7天,吸光度值与阴性对照组接近。结论:蜂胶对人牙龈成纤维细胞的毒性较小,不会增加奥硝唑的细胞毒性,并且毒性作用是可逆的;蜂胶及自制蜂胶奥硝唑合剂作为牙周病局部用药,具有进一步的研究价值。 Objective: To evaluate the cytotoxicity of propolis and propolis-ornidazole mixture on human gingival fibroblasts and provide information about local application of propolis and propolis-ornidazole mixture on periodontal diseases. Methods: Human gingival fibroblasts (HGFs) were cultured in medium containing propolis, ornidazole or propolis-ornidazole mixture at different concentrations for 24 h and the relative growth rates(RGR) were determined by MTT assay. Then the cells were cultured for 7 more days in medium without added materials and the optical density value was determined every day. Results :The RGR of propolis group with concentration of 0. 5 g/L was 93.9 percent. The average optical density values of two mixture groups were higher than ornidazole group and there were statistical differences between three groups when the concentration of ornidazole was 0.4 g/L or 0.8 g/L( P 〈 0.05 ). HGF of the experimental groups kept RGR increasing after the original 24 h culture. The optical density values of experimental groups showed no obvious differences when compared to control group after 7 days. Conclusion: Both of propolis and propolis-ornidazole mixture showed no cytotoxicity on HGF. Local application of propolis and propolis-ornidazole for treating periodontal diseases is feasible.
