摘要
目的建立大鼠血浆中替尼泊苷的HPLC方法。方法采用HPLC法,使用反相C_(18)柱(250mm×4.60mm,5μm,Phe- nomenex),流动相为甲醇:水:醋酸(38:60:2,V/V),流速为1.0 mL·min^(-1),检测波长230nm。结果替尼泊苷为0.5~25.0mg·L^(-1)时呈线性关系(r=0.9999),血浆检测限为0.06mg·L^(-1)。日内、日间RSD均<5%。给药后30min血浆的质量浓度为(4.21±0.37)mg·L^(-1)(n=4)。结论本方法简便、灵敏,可用于体内测定替尼泊苷。
AIM To improve a HPLC-UV method for the determination of teniposide in rat plasma.METHODS The concentration of teniposide was determined by using HPLC-UV method with the RP-C18column(250 mm×4.60 mm, 5 tan,Phenomenex).The mobile phase consisted of methanol-distilled water-acetic acid(38:60:2,V/V)with a flow rate of 1.0 mL·min^-1,whileγ= 230 nm.RESULTS The linear range was 0.5-25.0 mg·L^-1(r=0.9999).The limit of detection was 0.06mg·L^-1.RSD of inter-day and intra-day were all less than 5%.The concentration of teniposide in rat plasma at 30 min time point was (4.21±0.37)mg·L^-1(n = 4)after intravenous administration of teniposide solution.CONCLUSION The analytical method in this study is simple,sensitive,accurate,which can be easily applied to teniposide pharmacokinetic and pharmacological studies.
