摘要
目的探讨超声破坏微泡造影剂刺激大鼠心肌血管内皮生长因子(VEGF)分泌、促进新血管生成的作用。方法建立大鼠心肌梗死模型,随机分为4组。A组行超声破坏微泡治疗,B组行单纯超声治疗,c组单纯注射微泡,D组注射生理盐水作为对照,每组均处理3次。4周后取材,免疫组化检测心肌组织中VEGF、CD34表达,ELISA及Western blot检测VEGF表达。结果A组心肌组织中见大量VEGF和CD34表达,新生血管较多;B组有较少VEGF和CD34表达,新生血管较少;C组和D组仅有极少量VEGF和CD34表达,未见明显新生血管。ELISA检测显示,A组VEGF表达量明显增高[(8.194±1.129)pg/g],与其他三组[B组(4.339±0.753)pg/g,C组(2.304±0.762)pg/g,D组(2.411±0.801)Pg/g]比较,差异均有统计学意义(P〈0.05)。Westernblot检测显示A组有一条约21kD的明显的蛋白质条带表达。结论超声破坏微泡可刺激心肌内源性VEGF分泌,促进血管新生。
Objective To investigate whether ultrasound-mediated microbubble destruction can stimulate secretion of VEGF and angiogenesis in myocardium of rat. Methods Forty normal Wistar rats were randomly devided into 4 groups,each group was successed raised myocardial infarction model. In group A, the myocardium was exposed to ultrasound after intravenous injection of microbubble of 1.0 ml; in group B, ultrasound was simply used in the myocardium;in group C, microbubble was intravenous injection only; in the control group (group D), normal saline was infused. Each group was received 3 experiments. Immunohistochemical staining of VEGF and CD34 were performed, and the quantitate of VEGF was detected by ELISA 4 weeks later. Western blot was performed for expression of VEGF. Results Immunohistochemical staining showed that the expressions of VEGF and CD34 were higher than those in group B, C, D;and the new vessels degeneration were significant observed in group A than those in group B, C, D. ELISA detection showed that the quantities of VEGF were siginificant different in groups( P 〈0.05), it was highest in group A. A protein strap about 21 kD was shown in group A by Western blot detection,a tenuous protein strap about 21 kD was shown respectively in the other three groups. Conclusions Ultrasound-mediated microbubble destruction can facilitate the endogenous secretion of VEGF and accelerate the vascularization in the rat myocardium.
