热休克凋亡乳腺癌细胞负载的DC体内激发Th1型免疫反应 被引量:5

摘要 目的:探讨乳腺癌患者免疫热休克凋亡乳腺癌细胞负载的树突状细胞(DC)后体内免疫状态的变化,并对其安全性进行初步评估。方法:分离患者外周血单核细胞,用重组人粒细胞-巨噬细胞集落刺激因子(rhGM-CSF)和白介素-4(rhIL-4)诱导DC;将患者自体肿瘤细胞热休克后诱导其凋亡并负载自体DC。对10例确诊为乳腺癌的患者进行1个疗程(4次)的DC免疫治疗。结果:治疗中未出现明显不良反应,生活质量均有不同程度的改善,血清中IL-2、IL-12、IFN-γ和TNF-α水平治疗后呈明显上升趋势。结论:热休克凋亡乳腺癌细胞负载的DC体内可激发Th1型免疫应答,增强机体抗瘤能力,同时无明显毒副反应。
