摘要
在担子菌中,编码线粒体中间肽酶(mitochondrial intermediate peptidase,MIP)的基因普遍与交配型A因子紧密连锁。通过mip基因来定位和分离交配型A因子将是一种有效的研究食用真菌交配型的途径。本研究通过简并PCR法克隆得到一段531bp的香菇mip基因片段。经序列比对分析,该序列与已知的担子菌的mip基因之间有很高的同源性。
A 531 bp fragment of a putative mitochondrial intermediate peptidase (mip) gene from Lentinula edodes was amplified by PCR using a pair of degenerate primers designed according to conserved sequences of mip genes reported in other Basidiomycetes. Alignment analysis revealed that this putative partial mip gene from L. edodes exhibited high homology with corresponding segments of mip genes from the other Basidiomycetes. Since the gene encoding MIP appears to be very closely linked with the A type mating gene of Basidiomycetes, identification of mip may prove a useful tool for locating and cloning the A type mating gene in L. edodes and other edible fungi.
