耳针对血管性痴呆大鼠学习记忆障碍及NMDAR1的影响 被引量:5

摘要 目的:观察耳针对血管性痴呆(VD)模型大鼠学习记忆能力及脑内N-甲基-D-天门冬氨酸受体1(NMDAR1)的影响。方法:采用4-血管阻断(4-VO)的方法,复制大鼠血管性痴呆模型,采用Y-型迷宫,进行行为学检测,定量测定其学习记忆成绩,用免疫组化法检测脑内NMDAR1表达水平的变化。结果:正常对照组脑组织有少量的NMDAR1免疫阳性神经细胞分布,VD模型组NMDAR1免疫阳性细胞数较正常对照组显著增高,平均光密度显著降低(P<0.05);耳针治疗组脑、肾两耳穴交替治疗3周脑组织NMDAR1免疫阳性神经细胞数显著减少,平均光密度值增高(P<0.05)。结论:耳针对脑缺血性神经元损伤的保护作用可能是通过抑制NMDAR1的过表达、下调NMDAR1的数量而实现的,从而改善VD大鼠学习记忆障碍。 Objective: To observe the effect of auricular needling on abihties of learning and memory as well as expression of N MDAR1 in rats with vascular dementia(VD). Methods: VD rat model was established by blocking 4- vessel. The immunohistochemistry method was used to detect the changes of NMDAR1 expression, the Y-type maze test was used to measure the abilities of learning and memory. Results: There were slight expression of N MDAR1cells in the brain tissue of normal group. Compared with the normal group , the expression of N MDAR1 positive cells in model group were significantly enhanced, but the average optical density is obvious reduced( P 〈 0.05). After the treatment of auricular needling, the expression of NMDAR1 in model group was obviously reduced, and the average optical density was increased( P 〈 0.05). Conclusion: Auricular needling could ameliorate the cerebral ischemia-induced neuron injury by refraining overexpression of NMDARI, so as to improve the learning and memory of VD rats.
