摘要
目的研究葛根素在乙醇/硫酸铵两水相体系中的分配特性及其影响因素。方法变化体系中乙醇的浓度、添加不同分子量及浓度的PEG、改变体系pH值。结果葛根素的分配系数与回收率和体系中乙醇的浓度成正比,小分子量的PEG更有利于葛根素在上相的分配,增加pH值,对提高葛根素的分配系数有正面影响。结论最佳分配体系是向组成为乙醇(21%)/硫酸铵(20%)的体系中添加5%(W/W)的PEG400,最佳萃取环境为pH=7.8。
Objective To study the partition properties and influential factors of puerafin in aqueous two - phase system of ethanol/ammonium sulfate. Methods Change the concentration of ethanol in the system , add PEG with different molar mass and concentration, valuate pH. Results Partition coefficient and extraction ratio of puerarin increased with the increase of ethanol , PEG with low molar mass could facilitate puerarin to accumulate in the top phase , and the increased pH was more advantaged to the increase of the partition coefficient of puerarin . Conclusion Ethanol (21%)/Ammonium Sulfate (20%) with 5% (W/W) PEG400 is the improved extraction system, and the optimum extraction pH is 7.8%.
