摘要 目的:研究姜黄素对人前列腺增生细胞BPH1增殖及基因表达的抑制。方法:MTT法测定姜黄素抗增殖效应;逆转录-聚合酶链反应(RT-PCR)技术检测细胞凋亡抑制基因-2(Bcl-2)和增殖细胞核抗原(PCNA)mRNA的表达。结果:各实验组的姜黄素对增生细胞的生长均有抑制,抑制作用具有时间/剂量依赖性;姜黄素作用于BPH1细胞24h,Bcl-2和PCNA mRNA的表达水平下降,呈剂量依赖性。结论:姜黄素可抑制人良性前列腺增生细胞BPH1的增殖,其机制可能与姜黄素下调Bcl-2和PCNA mRNA的表达有关。 OBJECTIVE: To investigate the inhibitory effect of Curcumine on the proliferation and gene expression of human prostatic hyperplasia cell line BPH1. METHODS: MTT were used to examine the cell growth of Human prostatic hyperplasia cell line BPH1 after treated with different doses of Curcumine. The expressions of PCNA mRNA and Bcl - 2 at transcription were detected by RT-PCR. RESULTS: Curcumine inhibited cell growth time or dose- dependently and downregulated the expressions of PCNA and Bcl- 2 in BPH1 cells dose-dependently within 24 h after being treated by Curcumin.CONCLUSION: The antiproliferative effect of Curcumine on BPH1 might be associated to the downregulation of PCNA and Bcl - 2 levels.
