分化抑制蛋白Id1和PCNA及Ki67在肝细胞癌中的表达及意义 被引量:8

摘要 目的研究肝细胞癌中Id1表达与PCNA、Ki-67的关系及其临床意义。方法选本科存档标本肝癌20例、肝硬化20例和正常肝20例,免疫组化染色检测Id1、PCNA和Ki-67供研究。结果Id1:肝细胞癌,1分2例(10.0%),2分7例(35.0%),3分11例(55.0%);肝硬化,1分14例(70.0%),2分6例(30.0%);正常肝,1分17例(85.0%),2分3例(15.0%,P<0.01)。PNCA:肝细胞癌1分1例(5.0%),2分2例(10.0%),3分17例(85.0%);正常肝1分6例(30.0%),2分2例(10.0%),3分12例(60.0%,P>0.05)。Ki-67:肝细胞癌1分11例(55.0%),2分7例(35.0%)和3分2例(10%);肝硬化,0分6例(30.0%),1分14例(70.0%),2分0例,3分0例;正常肝,1分18例(90.0%),2分2例(10.0%),3分0例(P<0.01)。结论肝癌Id1表达增强与Ki-67表达有相关性(P<0.01),PNCA与Id1表达不一致,Id1可作为肝细胞增殖标志物。 Objective To study the expression of Id-1 in hepatocellular carcinoma and its correlation with PCNA and Ki-57 and its clinical significance. Methods The specimens of 20 cases of hepatocellu- lar carcinomas(HCC) ,20 cases of cirrhosis of liver and 20 cases of normal liver tissue were collected from files of specimens in our department. The specimens were examined by immunohistochemical stain with Id-1, PCNA and Ki-67 for study. The results of cytoplasmic Id-1 and nuclear staining of PCNA and Ki-67 were de- termined independently by observers. Results The percentage of positive cells and staining intensity of Id-1 were as follows : HCC, 1 point in 2 cases ( 10.0% ), 2 points in 7 ( 35.0% ), and 3 points in 11 ( 55.0% ) ; cirrhosis of liver(CL), 1 point in 14 cases(70.0% ), 2 points in 6(30.0% );normal liver tissue (NLT), 1 point in 17 cases(85.0% ), 2 points in 3( 15.0% ,P 〈 0.01 ). The rates of PCNA were as follows :HCC, 1 point in 1 case(5.0% ) ,2 points in 2( 10.0% ) ,and 3 points in 17(85.0% ) ;CL, 1 point in 8 (40.0%) ,2 points 0,and 3 points in 12(60.0% ) ;NLT, 1 points in 6 cases(30.0% ) ,2 points in 2( 10.0% ) ,and 3 points in 12 (60.0%, P 〉 0.05 ). The rates of Ki-67 were as follows : HCC, 1 point in 11 ( 55.0% ), 2 points in 7 (35.0%) ,and 3 points in 2 ( 10.0% ) ; CL,0 point in 6 ( 30.0% ), 1 point in 14 ( 70.0% ), 2 points 0 and 3 points 0;NLT,1 point in 18(90.0% ) ,2 points in 2( 10.0% ) ,and 3 pionts 0( P 〈 0.01 ). Conclusions The study showed that Id-1 geue expression increased in HCC, and correlated with the expression of Ki-67 (P 〈 0.01 ). PCNA as a marker of proliferation did not identical with Id-1 ( P 〉 0.05 ). Id-1 gene may serve as a proliferative marker of HCC.
