高强度军事训练致急性肾损伤动物模型的研究 被引量:15

摘要 目的建立高强度军事训练致非创伤性急性肾损伤的大鼠模型,为军事训练伤的研究提供技术平台。方法将40只12周龄的SD大鼠随机分为对照组(n=8),普通运动组(n=16)、军事训练组(n=16),按不同方法行一次性力竭跑台训练,观察训练结束即刻及恢复24h后血、尿指标及肾组织病理学改变情况。结果军事训练组大鼠训练结束即刻和恢复24h后尿总蛋白、白蛋白、N-乙酰-β-D-葡萄糖苷酶(NAG酶)、血清尿素氮、肌酐及血浆、肾组织血管紧张素Ⅱ(AngⅡ)浓度均高于对照组和普通运动组(P<0.05)。军事训练后24h,可见大鼠部分肾小管、肾小囊轻度扩张,小管上皮细胞轻度肿胀。结论该模型出现了肾损伤的临床特征,符合军事训练特点,可行性、重复性好,适用于高强度军训致急性肾损伤的基础和临床研究。 Objective To reproduce an animal model simulating acute kidney injury caused high intensity military training in rats by forced exhausting running on the treadmill,that could be used for the investigation of problems arising from military training.Methods 40 male SD rats were randomly divided into three groups:control group(n=8),ordinary exercise group(n=16)and forced exercise group(n=16).The animals in ordinary exercise group were commonly fed.The animals in the other two groups were forced to take on one-time exhausting running exercises on the treadmill by different ways.The rats in forced exercise group were forced to run on the treadmill till exhaustion to reproduce the animal model of overtraining.The indexes of urine,serum and kidney tissues were observed and analyzed shortly after training and 24 hours later.Results After training,the values of urine protein,urinary albumin,urease,serum urea nitrogen,creatinine,angiotensin Ⅱ in serum and kidney tissues of the animals in forced exercise group increased obviously compared with those in control group and ordinary exercise group.Those indexes in ordinary exercise group were higher than in control group(P〈0.05).24 hours later,those indexes of the animals in forced exercise group were still higher than that of the other two groups(P〈0.05).A gentle ectasia of renal tubule and capsule combined with cellular swelling and vacuolar degeneration were observed after 24 hours training in the rats of forced exercise group.Conclusions The animal model established is stable,reliable and repeatable.It can display the clinical features of acute kidney injury,and coincide with feature of military overtraining.It is used for scientific research of the acute kidney injury caused by military overtraining.
