摘要
目的:探讨抑制库普弗细胞对急性肝衰竭模型鼠Toll样受体4(Toll-like receptor4,TLR4)表达的影响。方法:80只SD雄性大鼠随机分为正常对照组、氯化钆(gadolinium chloride,GdCl3)组、内毒素(LPS)组、GdCl3+LPS组。免疫组化检测ED1表达,观察GdCl3对库普弗细胞的抑制程度,并检测血清ALT、TNFα、IL-1β的水平,Westernblot分析肝组织TLR4的表达。结果:GdCl3组肝组织中ED1免疫反应阳性细胞与正常对照组相比明显减少(P<0.05),LPS组ED1阳性细胞数量最多,GdCl3+LPS组ED1阳性细胞数少于LPS组(P<0.05)。GdCl3+LPS组ALT、TNFα、IL-1β水平高于正常对照组,但低于LPS组(P<0.05)。GdCl3+LPS组大鼠肝脏TLR4表达与LPS组相比较弱(P<0.05),与正常对照组相比差异无统计学意义(P>0.05)。结论:抑制库普弗细胞激活可减少肝组织TLR4的表达,库普弗细胞在急性肝衰竭中发挥重要作用。
Objective To investigate the effects of Kupffer cell inhihition on the expression of TLR4 in the rats with acute liver failure. Methods Eighty male SD rats were randomly divided into four groups as follows : normal control group, gadolinium chloride (GdCl3)group, LPS group and GdCl3 +LPS group. To identify the degree of Kupffer cell inhihition hy GdCl3, ED1 expression in liver tissue was observed by immunohistochemistry, and TLR4 expression was detected by Western blot. Serum ALT, TNFα and IL-1β levels were measured. Results The number of ED1 positive cells in liver tissues of GdCl3 group was lower significantly than that in control group (P 〈 0.05). Moreover, the number of ED1 positive cells in LPS + GdCl3 group was lower than that in LPS group (P〈 0.05), which had the most ED1 positive cells. Serum ALT concentration was elevated in the D-Gal/LPS treated rats, which was offsetted by the use of GdCl3 pretreatment. Serum ALT concentration was not elevated in rats receiving either saline or GdCl3 alone. Similar results were observed with serum TNFα and IL-1β levels (P 〈 0.05). The expression of TLR4 in GdCl3 + LPS group was significantly lower than that of LPS group (P 〈 0.05), but had no statistical difference compared with control group (P 〉 0.05). Conclusion Inhibiting activation of Kupffer cells may decrease the expression of TLR4 in rat liver tissues, and Kupffer cells play an important role in acute liver failure.
