摘要 目的观察骨髓间充质干细胞(BMSC)的电生理特性,为临床干细胞移植的安全性提供参考。方法分离并培养BMSC,应用全细胞膜片钳技术记录BMSC的电生理特性。结果BMSC的静息膜电位为(-33.6±6.8)mV,平均细胞膜电容为(32.3±8.7)pF(n=30)。电压钳实验显示,BMSC上可记录到KCaI、to,IKSUS和IK14种通道电流,但未记录到INa和ICa通道电流。结论BMSC上存在4种不同类型钾离子通道。 Objective To observe the electrophysiological characteristics of the mesenchymal stem cells (BMSCs) and to provide the reference for the safety of BMSC transplantation. Methods BMSC was isolated from rat bone marrow and cultured, and the whole-cell patch clamp technique was used to record the resting membrane potential, action potential, membrane capacitance, and ionic currents in BMSC. Results The resting membrane potential of rat BMSC was ( - 33.6 ± 6.8) mV, and the membrane capacitance was (32.3 ± 8.7 ) pF. The voltage-clamp experiment showed that there were four types of channels existing in rat BMSCs. They were KCa, Ito, IKSUS, and IKI Additionally, INa and ICa were in the absence of BMSC. Condusion Four types of potassium channels exist in rat BMSCs.
