摘要 目的 建立全血中环孢素A药物浓度的HPLC测定方法。方法 以Nucleodur C18(150mm×4.6mm,5μm)为分析柱,乙腈-水(71:29,v/v)为流动相,柱温为65℃,检测波长为210nm。先以80%乙腈沉淀蛋白,沉淀后的上清液过SPE小柱处理后进样分析。结果 环孢素A在40.0~1600.0 ng·mL^-1线性关系良好(r=0.9998),在3个浓度下的平均回收率〉95.0%,日内、日间RSD<10.0%,环孢素A平均萃取回收率〉86.0%,环孢素D平均萃取回收率为91.2%。结论本方法稳定、准确,能用于测定环孢素A血药浓度。 Objective To establish an HPLC method for assay of cyclosporine A in whole blood. Methods The chromatographic column was Nucleodur C8 (150 mm× 4.6 mm, 5 μm), the mobile phase consisted of acetonitrile and water (71 : 29, v/v), the column temperature was controlled at 65 ℃ and the UV detector was operated at 210 nm. The whole boold samples were added the 80% acetonitrile precipitate albumen, after the SPE subminiature column was disposed , the sample was directly injected into the system. Results The calibration curve was linear in the range of 40. 0~1 600.0 ng· mL^-1. The average recoveries of the three concentrations were all higher than 95.0%. The intra-day RSD and inter-day RSD was both less than 10.0%. the extraction recovery of cyclosporine A was higher than 86. 0%. Conclusion The method is steady and accurate. It is suitable for measuring cyclosporine A in whole blood.
