摘要 摘要:为了分析B-藻红蛋白的三种亚基,采用反相HPLC并通过二极管阵列/紫外可见光检测器和荧光检测器,以及LC-电喷雾/MS质谱检测器进行在线检测与鉴定.结果表明,根据在线检测的光谱特征,α,β和3个明显的γ亚基都能成功地被C4柱分离,且α和β亚基均在555 nm处有最大吸收峰,而γ亚基在493nm和555nm处有最大吸收峰.并结合质谱信息,可鉴别出α和β亚基,它们的相对分子质量分别为18977和20330.说明结合上述几种检测技术可为B-藻红蛋白的亚基鉴定提供可靠的结果. In this paper, three kinds of subunits of B-phycoerythrin (B-PE) were tested and identified on line by using inverse HPLC with photodiode array/UV-Vis and fluorescence detector, and by using LC-electrospray ionization/MS. The results show that the α andβ subunits as well as three y subunits of B-PE are all successfully separated by C4 column according to the on-line spectroscopic characteristics, and that both the α and the β subunits have a maximum absorption at 555 nm, while the γ subunit species show their maximum absorptions at 493 and 555 nm. Moreover, by coupling with the MS, α andβ subunits with respective relative molecular masses of 18977 and 20330 can be identified. It is thus concluded that a combination of the techniques mentioned above helps to identify the subunits of B-PE reliably.
