摘要 目的:研究高密度壳聚糖(high density chitosan,HDC)对小鼠L-929细胞的毒性,以期为其临床安全应用提供依据。方法:观察不同浓度HDC浸提液对体外培养L-929细胞形态的影响,四甲基偶氮唑蓝(methlthiazolyl tetrazolium,MTT)实验评价HDC对L-929细胞生长和增殖的影响,流式细胞术(flow cytometry,FCM)检测HDC浸提液对L-929细胞凋亡的影响,单细胞凝胶电泳(single cell gel electrophoresis,SCGE)检测不同浓度HDC浸提液对培养的L-929细胞DNA分子损伤的影响。结果:HDC浸提液对体外培养的L-929细胞形态无明显影响,对细胞生长和增殖无明显抑制作用,不同浓度材料浸提液的细胞毒性均为0-1级;FCM示凋亡率随浸提液浓度升高而升高,但不明显;SCGE结果显示HDC对L-929细胞的DNA无明显损伤作用。结论:HDC具有良好的细胞相容性和分子相容性,可为临床的安全应用提供依据。 Objective To study the toxicity of high density chitosan(HDC) in L-929 cell for its safe useness in clinic. Methods The morphological changes of L-929 cell was observed in vitro by invert microscope. MTT( methlthiazolyl tetrazolium) assay was used to evaluate the growth and proliferation of L-929 cell, the apoptosis rate of L-929 cell was detected by flow cytometry(FCM),the effects on DNA damage of cultured L-929 cell were observed through SCGE. Resuits The results of MTT showed that a series of concentrations of leaching liquors of HDC had neither reliable effect on cell morphology, nor significant inhibition on cell growth and proliferation. The cytotoxicity gradation of various concentrations of the extract of HDC ranged from 0 to 1 degree, the same results were discovered of the morphological change by invert microscopy. Apoptosis rate went up gradually with the increase of concentration of the extract liquid of HDC, but not significantly. There was no remarkable damage in comet rate to L-929 cells in DNA level. Conclusion HDC, having good cytocompatibility and molecular biocompatibility, might be used safely in clinlic.
