Expression of AQP4 in human menlngioma and the biological significance

摘要 目的研究水通道蛋白4(aquaporin 4 AQP4)在人脑膜瘤中的表达与脑膜瘤的病理学分型的相关性,并探讨其与瘤周脑组织水肿的关系及其可能机制。方法取伴水肿(MRI证实)的良性脑膜瘤组织15例,无水肿良性脑膜瘤组织21例,恶性脑膜瘤15例和正常脑组织6例,应用免疫组织化学方法检测AQP4在脑组织的表达,并结合临床病理参数进行综合分析。结果在不伴脑水肿的良性脑膜瘤组、伴水肿的良性脑膜瘤组以及恶性脑膜瘤组中阳性表达率均升高,正常脑组织中几乎未见阳性表达。伴水肿的良性脑膜瘤组织中AQP4阳性表达率高于无水肿良性脑膜瘤组(P〈0.05),恶性脑膜瘤组织中AQP4阳性表达率明显高于不伴脑水肿良性组(P〈0.01),AQP4的表达和脑膜瘤病理分级无相关性(P〉0.05)。AQP4的表达与瘤周水肿显著相关(P〈0.05)。结论AQP4在脑膜瘤中的高表达与脑组织水肿的发生密切相关。检测脑膜瘤AQP4的表达对了解脑膜瘤生物学行为和判断预后有重要价值。 Objective To investigate the correlation between the expression of AQP4 in meningioma and the clinicopathologic classification of meningioma, and explore its relationship with peritumoral edema and the mechanism. Methods The expression of AQP4 in 57 cases of meningiomas including 15 cases of benign meningiomas with severe perilesional edema on MRI scans,21 cases of benign meningiomas without edema, 15 cases of malignant meningiomas and 6 cases of normal brain tissues was detected by immunohistochemistry SP methods, The cllnicopathological parameters were analyzed at the same time. Resuits No expression of AQP4 was observed in normal tissues. It was significant difference in the expression of AQP4 between the cases with severe perilesional edema and those without edema ( P 〈 0.05 ), and between the malignant meningiomas and the benign meningiomas without edema (P 〈 0.01 ). The expression of AQP4 had no correlation with pathological grade ( P 〉 0.05 ). The expression of AQP4 was positively correlated with peritumoral edema ( P 〈 0.05). Conclusion In meningioma, AQP4 may play an important role during malignancy conversion and peritumoral edema formation. Detection of the expression of AQP4 may be of value in understanding the biological behavior and predicting the prognosis of meningioma.
