摘要
选用了29份小麦材料,研究了变性剂SDS对小麦PPO活性的影响,深入了解了小麦PPO的生化特征,建立了一种更有利于筛选不同PPO活性品种的检测方法,用来辅助小麦面制食品颜色性状的遗传改良。结果表明,SDS可以增加小麦PPO活性,50mmol/L的SDS是提取小麦PPO的最适浓度。用SDS提取时,PPO活性很稳定。SDS不但可以增加小麦PPO的提取效率,而且可以诱导酶结构发生变化,对酶活性产生影响,对高活性品种有激活作用,对低活性品种有抑制作用,对提取效率的影响大于对酶活性的影响。加入SDS后提取检测的全粉、小麦粉、籽粒PPO活性与面片24hL*值及0~24hL*值的变化显著相关。所以用sDS提取检测小麦PPO活性更有利于筛选不同PPO活性的种质,尤其是提高筛选低PPO活性种质的可靠性。此外,小麦PPO在加入SDS后所呈现的生化特征为小麦体内PPO生理机制的完善提供了理论依据。
29 wheat samples were used to study the influence of detergent SDS on wheat polyphenol oxidase (PPO) activity and to develop a more efficient assay method for PPO activity, which would be useful in heredity improvement of the color characteristic of wheat - based products. Results indicate that SDS can enhance the wheat PPO activity and 50 mmol/ L is the optimum SDS concentration for the wheat PPO extraction. The PPO activity is quite stable in presence of SDS. SDS not only enhances the extraction efficiency of the wheat PPO, but also activates the PPO from high - PPO lines and inhibits the PPO from low- PPO lines by inducing conformational change. The PPO activity assayed after extraction in presence of SDS shows significantly negative correlation with the dough sheet brightness ( L*) at 24th h and highly significant positive correlation with the change in dough sheet brightness during 0 - 24 h. So the PPO activity assayed after extraction in presence of SDS is more reliable for screening the wheat germplasm with different PPO activity, especially for low PPO activity wheat germplasm. Moreover, the biochemical properties of PPO present in the process can offer theoretic evidences for the integrity of PPO physiological mechanism in wheat.
