HER-2癌基因反义寡核苷酸对乳腺癌细胞化疗敏感性的影响

Effects of HER-2 oncogene antisense oligodeoxynucleotides-folate on chemosensitivity in breast cancer cell line in vitro

摘要 目的研究HER-2癌基因反义寡脱氧核苷酸联合叶酸对乳腺癌细胞SK-Br-3化疗敏感性的影响。方法通过叶酸受体将HER-2反义寡脱氧核苷酸导入乳腺癌细胞,给予不同剂量的阿霉素和顺铂,用四氮唑盐(MTT)方法检测细胞存活率。结果反义寡脱氧核苷酸-叶酸(ASODN-FA)显著增加SK-Br-3细胞对阿霉素和顺铂的敏感性,使细胞存活率明显降低。结论HER-2反义寡脱氧核苷酸能增加乳腺癌细胞对阿霉素和顺铂的化疗敏感性。 Objective To study the effects of HER-2 oneogene antisense oligodeoxynudeotide-folate ASODN-FA HER-2 ASODNs , as a oncogene inhibitor, on chemotherapeutic drug sensitivity in human breast cancer cells line SK-Br-3. Methods Human breast cancer cells were cultured and inoculated into the wells, then added ASODN-FA 100 μl. The chemotherapeutic drug adriamycin(ADM) and cisplatin (DDP) were administrated different concentration to observe the effect of ASODN on the cell survival rate by MTT. Results ASON-FA HER-2 ASODNs inhibited the expression of HER-2 onco gene which significantly increased the chemosensitivity of breast cancer cells SK-Br-3 on ADM and DDP. Conclusion HER-2 antisense oligodeoxynucleotide can effectively enhance the sensitivity of human breast carcinoma cells to the chemotherapeuties drug adfiamycin and cisplatin.
