摘要
尝试了固体碘熏蒸显示透明圈,然后直接传代培养菌株检测耐受时间及存活后产异淀粉酶的栖热菌酶活变化,同时与碘液染色及碘液熏蒸进行对比。结果显示用0.04 g碘固体熏蒸1 m in效果较好,直接传代菌落存活率高,对生产性能没有显著影响,不易污染杂菌,方便、快捷,可替代碘液显色法。
Iodine vapor was used to visualize halos for screening of extracellular amylolytic enzyme produ- cing strains. The screened colonies were directly transferred to test the persistent time and isoamylase ac- tivity of Thermus sp. Lugol's iodine solution coloration was compared with Lugol's iodine vapor method in the screening. Experimental results showed that vapor with 0. 04 g iodine for 1 minutes was preferred con- dition of efficient and harmless to the strains and the production traits.
