p53在神经酰胺诱导内皮细胞凋亡中的作用 被引量:2

摘要 目的:探讨p53在外源性神经酰胺(C2-ceramide)诱导人脐静脉内皮细胞凋亡中的作用。方法:体外培养脐静脉内皮细胞,用不同浓度的C2-ceramide进行处理,Tunel检测细胞凋亡;逆转录-聚合酶链反应(RT-PCR)和Western blot检测p53的表达;并用p53的抑制剂PFT-α(pifithrin alpha,PFT-α)预处理细胞,再加入C2-ceramide与对照组比较观察细胞凋亡指数。结果:C2-ceramide呈剂量依赖性诱导内皮细胞凋亡;p53的表达随着C2-ceramide浓度的增加呈现降低的趋势;PFT-α不能抑制C2-ceramide诱导的凋亡。结论:神经酰胺能诱导内皮细胞的凋亡,可能不依赖于p53途径。 Objective:To explore the role of p53 in ceramide-induced endothelial cells apoptosis. Method:Human umbilical vein endothelial cells(HUVEC) were treated with different dose of C2-ceramide;And endothelial cells were primarily cultured and treated with C2-ceramide alone or in combination with PFT-α(an inhibitor of p53) respectively. The apoptosis of cells was detected with Tunel. The expression of p53 protein and mRNA levels was measured by Western blot and reverse transcription polymerase chain reaction (RT-PCR). Result:C2-ceramide induces HUVEC apoptosis in a dose-dependent manner. Ceramide decreased the expressions of p53. PFT-α can not inhibit the HUVEC apoptosis induced by C2-ceramide. Conclusion:Ceramide-induced endothelial cells apoptosis is independent on p53-independent pathway.
