摘要
目的:建立HPLC法测定打碗花根中莨菪亭含量的方法。方法:采用Hypersil ODS2 C18色谱柱(4.6mm×250mm,5μm),流动相为乙腈1%冰醋酸水溶液(20:80),检测波长为341nm。结果:在1.07~21.4mg·L^-1范围内莨菪亭呈现良好的线性关系(r=0.9999);平均回收率为99.2%,RSD为1.06%。结论:用本法测定打碗花根中莨菪亭含量灵敏准确,为其质量评价提供了一定的参考依据。
OBJECTIVE To develop a HPLC method to determine the content of scopoletin in root of Calystegia hedracea Wall. METHODS The separation was accomplished on a Hypersil ODS2 C18 column(4. 6 mm × 250 mm,5 μm)with a mixture of acetonitrile - 1% aqueous acetic acid(20: 80)as the mobile phase. The detecting wavelength was 341 nm. RESULTS The linear range was 1.07- 21.4μg·mL^-1 (r= 0. 999 9) ;The average recovery of the assay was 99. 2%, RSD= 1.06%. CONCLUSION This method had adequate sensitivity and accuracy to measure the concents of scopoletin and could be used in the quality control in the root of calystegia hedrecea Wall.
