摘要 目的研究木瓜超氧化物歧化酶粗提物颗粒的加工工艺,并评价其生物学效应。方法1%纤维素酶酶解木瓜匀浆,β-环糊精和γ-环糊精包埋,制得超氧化物歧化酶(superoxide dismutase,SOD)粗提物,50只小鼠分为5组:空白对照组、模型对照组、3个试验组(样品剂量分别为1000、2000、3000U/kg)。对照组予同质量基础饲料,除空白对照组外,各组给予5Gy60Coγ射线全身照射1次,3d后处死,测定血清和各组织MDA含量及SOD活性。结果酶解后单位体积的上清液中SOD的活力比酶解前降低了7.8%,但是由于出汁率大大提高,使得单位质量木瓜所得上清液中SOD总活力提高近10.65%。采用3%的γ-环糊精包埋木瓜SOD,并运用喷雾造粒技术制备PC-SOD,其SOD活性保留率为67.8%。动物实验显示:各剂量组中小鼠血清和组织中的MDA含量均低于模型对照组,各剂量组中小鼠血清和组织中的SOD活性均高于模型对照组。结论木瓜的本方法SOD粗提物颗粒(PC-SOD)的加工工艺效果好。产品PC-SOD对降低小鼠血清和组织中的MDA有显著作用,且对提高小鼠血清和组织中的SOD活性也有明显的效果。 Objective To establish the technics of preparing the particle product of Chaenomeles chinensis superoxide dismutase extract (PC-SOD) and investigate the biological function of PC-SOD. Methods Enzymatic technique was applied to improve extraction rate. The activity of SOD during the process of preparing PCSOD was explored by adopting γ-dextrin and G-dextrin for packing. The biological function of PC-SOD was studied by detecting MDA contents and SOD activity in blood serum, liver and kidney of mice treated with PC- SOD product. Results In enzymatic process, juice recovery was higher and the total SOD activity was 10. 65% higher than non-enzymatic procedure. Using 3% γ-dextrin to pack SOD and prepare PC-SOD, the activity rate of SOD in PC-SOD was 67.8%. In mice fed with PC-SOD product, the MDA content in blood serum, liver and kidney was lower, while the SOD activity was higher than the control animals. Conclusion This technique of preparing the particle product of Chaenomeles chinensis PC-SOD is effective. PC-SOD reduces MDA content in blood serum and organs of mice, and increases the SOD activity.
