摘要
目的了解持续低剂量顺铂(DDP)联合α-干扰素(IFN-α)对小鼠移植性肝癌血管生成的影响。方法将48只接种H22肝癌的昆明小鼠随机分为4组,各12只。低剂量DDP组(DDP组):腹腔注射DDP 0.2 mL(0.6 mg/kg);IFN-α组(IFN组):皮下注射IFN-α0.2 mL(2×10^4U);低剂量DDP联合IFN-α组(联合组):腹腔注射DDP 0.2 mL(0.6 mg/kg)并皮下注射IFN-α0.2 mL(2×10^4U);生理盐水对照组(对照组):腹腔注射生理盐水0.2 mL,每天1次,连续2周。观察肿瘤体积、小鼠体质量的变化和药物毒副反应,采用免疫组织化学方法测定肿瘤微血管密度(MVD)、血管内皮生长因子(VEGF)的表达。结果DDP组和IFN组肿瘤生长比较缓慢,联合组抑瘤效果更为明显(F=260.58,q=0.72-1.08,P〈0.05),且毒、副反应未增加。与对照组比较,DDP组和INF组的MVD、VEGF表达均下降(F=157.04、52.97,q=9.55-19.02,P〈0.05),联合组下降更显著(q=5.25-9.47,P〈0.05)。结论低剂量DDP与IFN-α联合应用具有明显的抗血管生成协同作用,抑瘤效果显著,毒、副反应小。
Objective To evaluate the effect of combined low-dose cisplatin (DDP) with interferon-α (IFN-α) on angiogenesis in mice with planted hepatoma. Methods Forty-eight Kunming mice planted with H22 hepatoma were evenly randomized into four groups as DDP group: DDP 0.6 mg/kg,intraperitoneal injection; IFN group: IFN-α, 0.2 mL (2×10^4 U), hypodermic; combination group: DDP, 0.6 mg/kg, intraperitoneal, and IFN-α, 0. 2 mL (2×10^4 U), hypodermic; control group: normal saline, 0.2 mL, intraperitoneal. All injections were given once daily for two weeks. Volume and weight of the tumors, as well as weight of the mice were measured. The expression of microvessel density (MVD) and vascular endothelial growth factor (VEGF) was determined immunohistochemically. Results Tumor grew comparatively slow in DDP and IFN groups,and tumor inhibition was more significant in combination group without increase of any toxicity or adverse reactions (F= 260.58, q= 0. 72-1. 08, P( 0.05). Compared with the controls, MVD and VEGF expressions were lower in DDP and IFN groups (F= 157.04, 52.97;q= 9.55-19.02;P〈0.05), and more dramatic decline observed in combination group (q = 5.25- 9.47, P〈0. 05). Conclusion Low-dose DDP in combination with IFN-α has an obvious joint action of anti-αngiogenesis, with notable tumor inhibition and less side reactions.
