摘要
[目的]检测NDRG1基因在正常子宫内膜和Ⅰ型子宫内膜癌中的表达,探讨其在Ⅰ型子宫内膜癌发生中的作用机制。[方法]应用免疫组化、原位杂交及RT-PCR技术检测NDRG1基因在正常子宫内膜与Ⅰ型子宫内膜癌中的表达。[结果]免疫组化结果显示,NDRG1蛋白在正常子宫内膜和Ⅰ型子宫内膜癌中的表达率分别为12.5%和83.5%,有显著性差异(P=0.000);原位杂交结果显示,NDRG1mRNA在10例正常子宫内膜中只有2例表达,而在30例Ⅰ型子宫内膜癌中有22例表达,有显著性差异(P=0.007);RT-PCR结果显示,NDRG1mRNA在Ⅰ型子宫内膜癌中的表达比正常内膜增高3倍(P=0.000)。[结论]NDRG1在子宫内膜癌中在蛋白水平和mRNA水平与正常子宫内膜相比都明显升高。它可能是一种肿瘤相关基因,其异常表达可能参与了Ⅰ型子宫内膜癌的发病机制。
[Purpose] To explore the expression of NDRG1 in normal endometrium and endometriold adenocarcinoma type Ⅰ , and to investigate its carcinogenetic mechanism in endometrioid adenocarcinoma, [Methods] The expression of NDRG1 in normal endometrium and endometrioid adenocarcinoma were examined by immunohistochemistry, in situ hybridization and RT-PCR methods. [Results] The immunohistochemistry result indicated that the positive expression rate of NDRG1 protein in normal endometrium and endometrioid adenocareinoma was 12.5% and 83.5% respectively, with significant difference (P=0.000); NDRG1 mRNA expressed in 2 of 10 cases of normal endometrium and 22 of 30 cases with endometrioid adenocarcinoma by in situ hybridization (P= 0.007); In RT-PCR test, NDRG1 mRNA expression was 3 times higher in endometrioid adenocarcinoma than that of normal endometrium (P=0.000). [Conclusion] NDRG1 expression is upregulated significantly in endometrioid adenocarcinoma in contrast with normal endometrium both at protein and mRNA level (P〈0.01), NDRG1 might be a tumor related gene. Its overexpression may participate in the carcinogenesis of endometrioid adenocarcinoma.
