摘要 目的探讨微透析探针置入的刺激对大鼠纹状体内谷氨酸水平的影响。方法通过初次和二次置入微透析探针至大鼠纹状体并收集脑微透析液,结合高效液相色谱法检测透析液谷氨酸水平。结果探针初次置入纹状体谷氨酸水平立即升高约30倍,3h后降至基础值;二次置入时谷氨酸仅升高约15倍,1h后即降至基础值。结论微透析探针置入的刺激可引起大鼠纹状体内谷氨酸水平短暂增高,在脑微透析检测谷氨酸样品时应注意探针置入的影响。 Objective To investigate the influence of glutamate levels in rat striatum after intracerebral implantation of microdialysis probe. Methods Microdialysis probe was inserted into rat striatum by one-time or two-time insertions. Dialysate was collected and detected for glutamate levels with high performance liquid chromatograph(HPLC). Results The insertion of microdialysis probe into striatum at first time results in a transient raise in glutamate(about 30-fold above baseline), and declined to baseline within 3 hours. While glutamate levels were increased only 15-fold above baseline when microdialysis probe reinsert into striatum, and dropped to baseline within 1 hour. Conclusion The data suggest that the implant trauma may cause transient glutamate perturbations in rat striatum. Thus, the influence of probe implantation for detecting glutamate samples by. intracerebral microdialysis need to be considered.
