摘要
目的建立HPLC法同时测定当归四逆汤中4种有效成分的含量。方法采用HypersilODS色谱柱(250mm×4.6mm,5μm),以乙腈-体积分数为1.4%的冰醋酸水溶液进行线性梯度洗脱,流速:1.0mL·min^-1,检测波长:275nm,进样量:10μL。结果阿魏酸的线性为0.86~20.64mg·L^-1(r=0.9992),平均回收率为96.2%,RSD为1.5%;芍药苷的线性为20~200mg·L^-1(r=0.9993),平均回收率为97.3%,RSD为1.0%;肉桂酸的线性为2.1~27.3mg·L^-1(r=0.9995),平均回收率为94.0%,RSD为1.2%;甘草酸的线性为2.14~29.96mg·L^-1(r=0.9997),平均回收率为98.1%,RSD为2.0%。仪器与方法精密度均小于2.0%。结论该方法能更加全面控制该方的质量,优化后的提取工艺可为今后剂型的改进提供基础。
Objective To determine simultaneously the contents of four active components in Dangguisini decoction by RP-HPLC method. Methods The separation of four ingredients was achieved on a Hypersil C18 column (250 mm × 4.6 mm , 5μm) with gradient elution using solvents of acetonitrile and aqueous acetic acid. Results Assay was linear over the range 0.86 - 20.64 mg· L^- 1 for ferulic acid, 20 - 200 mg· L^- 1 for paeoniflorin, 2.1 - 27.3 mg· L^- 1 for cinnamic acid, 2.14 - 29.96 mg· L^- 1 for glycyrrhizin acid. Average recoveries of samples were all over 94.0 %, RSD were all below 2.0 %. Conclusions The method is simple and accurate and can be used as the method for quality assessment of Dangguisini decoction.
