摘要
目的:探讨阿托伐他汀对山羊静脉移植术后移植物内增厚程度的影响。方法:同品系山羊20只随机分为对照组和治疗组,每组10只动物。手术暴露双侧股动脉并部分切除造成动脉缺损。选取大隐静脉一段倒置吻合修复动脉缺损,治疗组在饲料中掺入阿托伐他汀片60mg.d-1,术后2周和4周取材,对移植静脉进行病理学、免疫组织化学检测,测定内膜厚度,PCNA指数和TUNEL指数,统计学分析各组间的差异。结果:形态学观察显示静脉移植术后2周和4周,两组内膜均不同程度的增厚。静脉移植术后4周,治疗组内膜厚度明显小于对照组[(194.51±17.02)vs(331.70±25.18)μm,P<0.01];术后4周治疗组PCNA指数明显少于对照组[(16.15±2.19)vs(28.89±2.24),P<0.01];术后4周,治疗组TUNEL指数明显大于对照组[(31.02±2.59)vs(20.74±1.86),P<0.01]。结论:阿托伐他汀治疗可以减轻山羊移植静脉内膜的增厚。
Objective: To determine whether atorvastatin inhibits intimaJ hyperplasia of vein graft in goats. Methods: Goats were randomly divided into the control and atorvastatin groups (n = 10 for each group). Arterial defects were made in bilateral femoral arteries; 4 cm vein grafts were removed from saphenous veins and transplanted to the arterial defects. The control goats were fed regular goat chow, while goats in atorvastatin group were fed atorvastatin (60 mg·d^-1 ) in regular chow. The vein grafts were harvested 2 weeks and 4 weeks after the operation. The thickness of the neointima was determined using a computerized image analysis system ; the intimal cells were evaluated by immunohistochemical staining for proliferative cell nuclear antigen (PCNA) and terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labelling (TUNEL). Results: The intimal thickness of grafts were significantly thinner in atorvastatin group than in control group [ (194.51 ± 17.02) vs (331.70 ± 25.18 )μm, P 〈0.01 ]. The numbers of PCNA-positive cells were less in atorvastatin group than in control group [ (16. 15 ± 2. 19) vs (28.89± 2.24), P 〈 0.01 ]. The numbers of TUNEL-positive cells were more in atorvastatin group than in control group [ (31.02 ± 2.59 ) vs (20.74 ± 1.86 ), P 〈 0.01 ]. Conclusion : Atorvastatin can inhibit intimal hyperpasia of vein graft in goats.
