摘要
对紫杉醇产生菌HD1-3的孢子分别进行了紫外线照射(UV)、亚硝基胍(NTG)和UV+NTG复合诱变处理,筛选制霉菌素抗性突变株。试验所确定的Nodulisporium sylviforme紫杉醇产生菌HD1-3孢子诱变的适宜条件为:将孢子悬液经过3.0mg·mL-1 NTG、处理20min后,在电磁搅拌下,用紫外灯(30w,距离30cm)照射处理45s。共筛选出17株正突变株。经发酵筛选试验,获得了一株遗传性状稳定、高产紫杉醇的诱变菌株——UN25-3,其紫杉醇产量从出发菌株的(232.73±4.61)μg·L-1提高至(327.14±7.63)μg·L-1。
In order to obtain resistant mutants to nystatin, ultraviolet radiation, NTG and UV + NTG were used to mutagenize the spores of taxol - producing fungus HD1-3, respectively. The spore suspension was treated with 3.0 mg ·mL^- 1 NTG for 20 min, followed by UV irradiation ( 30w, 30cm distance ) for 45 s under magnetic stirring. Seventeen strains were found to be positive mutants with high taxol when screened on plate containing 95 μg· L^-1 nystatin. After further screening experiments on fermentation, a mutant -UN25-3 which was able to produce taxol with high yield and could be stably passed on genetics was eventually found, it' s ability to produce taxol was improved from (232.73±4. 61) μg· L^-1(strain HD1-3)to (327.14±7.63) μg· L^-1(strain UN25-3).
