硒协同中药多糖对白血病细胞株P388体外作用的实验研究 被引量:5

摘要 目的:研究香菇多糖协同硒酸酯多糖对白血病细胞的抑制作用,探讨其相关机理。方法:MTT法检测香菇多糖协同硒酸酯多糖对白血病细胞增殖的抑制作用,流式细胞术检测细胞凋亡,免疫细胞化学检测相关基因Fas和FasL表达水平的改变。结果:香菇多糖和硒酸酯多糖可抑制白血病细胞增殖,硒酸酯多糖主要通过上调Fas和FasL促进凋亡发挥肿瘤抑制作用,香菇多糖主要通过上调Fas促进细胞凋亡,FasL表达没有变化。结论:二者存在不同抑癌机理。香菇多糖协同硒酸酯多糖可显著提高白血病抑制效应。 Objective: To study on the inhibitory effect of lentinan in combination with kappa- selenocarrageenan on leukemia and its molecular mechanism of antileukemia. Methods: The inhibitory effect of lentinan in combination with kappa - selenocarrageenan on leukemia cells was detected by MTT methods. The extent of cell apoptosis was measured by flow cytometry. The expressive levels of Fas and FasL were detected by immunochemistry assay. Results: Lentinan in combination with kappa- selenocarrageenan can inhibit the proliferation of leukemia cells. Kappa - selenocarrageenan plays tumor - suppressing roles through upregulating the expression of Fas and FasL and improving apoptosis, and lentinan by upregulating the expression of Fas but no effect on FasL.Conclusion: Lentinan and kappa - selenocarrageenan have different tumor - suppressing mechanisms. Lentinan in combination with kappa - selenocarrageenan can markedly improve the inhibiting effect on leukemia.
