摘要
以红斑大壁虎、黑斑大壁虎、蹼趾壁虎和中国石龙子为材料,采用改进的SDS/蛋白酶K裂解法提取DNA,用特异引物进行PCR扩增线粒体Cyt b基因部分序列,PCR产物经纯化后,进行序列测序。用邻接法和最大简约法构建了其系统发育关系,结果显示红斑大壁虎和黑斑大壁虎首先聚在一起,再先后与同为壁虎属的蹼趾壁虎和白脊壁虎相聚,然后与同为壁虎科的沙虎相聚,最后与石龙子科的中国石龙子相聚。
The Red Gekko gecko,Black Gekko gecko,Gekko subpalmatus and Eumeces chinensis being taken as materials,genomic DNA was extracted with the improved SDS/ proteinase K method,and specific primers were used for PCR to amplify the Cyt b gene sequences of mtDNA.Then the PCR products were purified and sequenced.Teratoscincus keyserlingii acting as out group,the phylogenetic trees was constructed with NJ and MP method.The results showed that Red Gekko gecko and Black Gekko gecko had the closest phylogenetic relationship,Gekko vittatus was followed by Gekko subpalmatus and the Teratoscincus keyserlingii was the farthest.
