不同野生稻居群DREB1B启动子的克隆与进化分析 被引量:1

摘要 根据Genbank所公布OsDREB1B的启动子序列设计引物,从12个不同普通野生稻居群和2个主要栽培稻亚种(籼、粳)克隆DREB1B启动子序列。结果表明,该启动子绝大部分区域都非常保守,存在1个AGC重复序列,若含有5个AGC重复,重复序列后第16个碱基为A,若含有6个或7个AGC重复,此位点碱基为T,并且在启动子中发现几个逆境相关的顺式作用元件。进化树分析表明,同一地域分布的居群存在相近的进化关系,但也受海拔等因素的影响。 Primer was designed according to OsDREB1B promoter sequence in the genbank. DREB1B promoter regions from 12 wild rice populations (Oryza rufipogon Griff) and 2 cuhivars (Japonica, Indica)were cloned and analyzed .The result showed that the promoter regions were conserved, including an AGC tandem repeated sequence. If there was five repeated AGCs,the 16th nucleotide after the repeated sequence was A,while it was T if the AGC repeats were more than 5. Meanwhile,there also existed some stress-related cis-acting elements in the promoters. Phylogenetic analysis showed that rice from the same geographical distribution had the close evolutionary distance, but it also affected by other factors, such as altitude.
