摘要
以4-甲基伞形酮磷酸酯为底物,利用动力学模式,建立了一种检测碱性磷酸酶活力的荧光法。确定了荧光检测的最佳条件为:激发波长为365nm,发射波长为449nm,作用时间为第2~6min,pH值为9,底物体积为100μL,底物缓冲溶液浓度为0.1mol·L^-1。该法操作简便、快速。
Taking 4-methylumbelliferyl phosphate liquid (4-MUP) as substrate, the fluorometric method of alkaline phosphatase (ALP)activity has been established with the kinetics pattern. The optimum detection conditions are determined as follows: the excitation wavelength is 365 nm and emission wavelength is 449 nm;the reaction time is 2th-6th min;the pH value is 9; substrate volume is 100μL, concentration of substrate buffer solution is 0. 1 mol · L^-1. The method is simple and fast.
