摘要
目的研究蛹虫草菌产胞外多糖的最佳发酵培养基和发酵条件。方法采用干重法测定菌丝生物量。采用硫酸-苯酚法测定多糖含量。结果蛹虫草产胞外多糖最佳发酵培养基组成为:蔗糖6%,硝酸钾0.2%,酵母膏2%,MgSO4.7H2O 0.02%,K2HPO40.04%,FeSO4.7H2O 0.002%;优化发酵培养条件为:接种量6%,初始pH7.0,温度24℃。结论在优化的培养基组成和发酵条件下,蛹虫草胞外多糖产量为4.84 g/L,菌丝体干重为21.12g/L。
Objective:To study the optimum media composition and fermentation conditions for exopolysaccharides production by Cordyceps militaris. Methods: Mycelium biomass was determined by dry-weight method. The levels of polysaccharides were determined by phenol-sulfuric acid method. Results: The optimum media composition was 6% sucrose, 0. 2% KNO3, 2% yeast extract, 0.02% MgSO4·7H2O, 0.04% K2HPO4 and 0.002% FeSO4·7H2O. The fermentation conditions were 6% of inoculation volume, initial pH7.0 and fermentation temperature 24℃. Conclusion: The yield of exopolysaccharides was 4.84 g/L and mycelium dry weight was 21.12g/L under the optimized conditions.
