摘要
目的:探讨采用不同分离方法所得青蒿粗提物应用后对结肠癌HT-29细胞活力的影响。方法:对青蒿水煎液分别用石油醚、氯仿、乙酸乙酯、正丁醇进行萃取及AB-8大孔树酯吸附后用蒸馏水、30%乙醇、50%乙醇、75%乙醇、95%乙醇进行洗脱柱层析,将分离得到的粗提物分别处理结肠癌HT-29细胞,采用MTT法对细胞活力进行检测。结果:每相粗提物浓度在200μg/mL范围内时,细胞活力下降程度较缓慢。在400μg/mL及600μg/mL应用后细胞活力与其他浓度比较均有明显降低(P<0.01)。结论:不同溶剂萃取及大孔树酯柱层析分离青蒿水煎液所得粗提物,应用浓度在200μg/mL内对HT-29细胞生长的抑制作用不明显,浓度大于400μg/mL后,细胞生长受到明显抑制。
Objective: To study the effects of different crude extracts of Abrotani herba on colon carcinoma HT - 29 cells. Methods: To extract the water decoctum of Abrotani herba using ligarine , chloroform, acetoacetate and n - butanol , and absorbed by macroporous resin AB -8 was eluted with distilled waterusing , 30% alcoh , 50% alcoh , 75% alcoh and 95% alcoh. Then detected the proliferation of HT - 29 cells using different crude extracts of Abrotani herba by MTT method. Results: In dose of infra - 200μg/mL , the proliferation of HT - 29 cells had little changes. In dose from 400 - 600 μg/mL , the proliferation of HT - 29 cells were inhibited obviously . Conchl^on: The inhibition of proliferation in HT - 29 cells were little in dose of infra - 200 μg/mL of different crude extracts of Abrotani herba , but in dose from 400 - 600μg/mL , the inhibition of proliferation in HT - 29 cells were obviously .
