摘要
目的:研究RCASl在纳米雄黄混悬液诱导人宫颈癌HeLa细胞凋亡中的表达,探讨其在细胞凋亡中的作用。方法:HeLa细胞经不同浓度纳米雄黄混悬液作用一定时间后,MTT法检测细胞生长、增殖状态,光镜及透射电镜下观察细胞形态及凋亡情况,ELISA检测RCA81蛋白量的表达,并进行图像分析。结果:纳米雄黄混悬液对HeLa细胞生长增殖有时间剂量依赖性抑制作用;25~50mg·L^-1纳米雄黄混悬液作用48~72h后,HeLa细胞出现凋亡细胞的形态学改变;ELISA检测表明HeLa细胞高表达RCASl蛋白,A值为(1.47±0.05),空白对照A值(0.139±0.015);25~50mg·L。。纳米雄黄混悬液作用48h后,RCASl蛋白的表达量A值分别降低为(0.423±0.021)和(0.146±0.018)(P〈0.01)。结论:纳米雄黄混悬液可抑制HeLa细胞的生长、增殖和诱导凋亡,并使RCAS1蛋白表达下降。
OBJECTIVE To study the expression and the effects of RCAS1 protein in apoptosis of HeLa cell induced by realgar nanometer suspension. METHODS After being incubated incuhure medium containing variable levels of realgar nanometer suspension, cell growth and proliferation of HeLa cell was analyzed with MTT assay; apoptosis was detected with cell morphology by light and transmission electron microscopy. The expression of RCASlprotein was determined with ELISA staining and imaginganalysis. RESULTS After the treatment, cell growth and proliferation of HeLa cell was inhibited in a dose-and time-dependent manner. Morphologically, after treatment with 25 to 50 mg·L^-1 of realgar nanometer suspension for 48 to 72 h, HeLa cells presented some features of apoptosis. The baseline RCASlexpression of HeLa cell was very high A: 1.47±0. 05 while the absorbance (opticaldensity) value(A) of blank control was 0. 139±0.015. The expression of RCAS1 protein in HeLa ceils co cultured with 25 to 50 mg·L^-1 of realgar nanometer suspension for 48 h A:0. 423±0. 021 and 0. 146± 0. 018 respec tively was decreased,compared with the untreated (P〈0.01). CONCLUSION Realgar nanometer suspension, could inhibit growth and proliferation of HeL,a cell and induce its apoptosis, and the down-regulation of RCAS1 protein.
