摘要
目的改进和完善大鼠急性心肌缺血再灌注损伤动物模型的制备,简化手术操作步骤,提高模型成功率和动物成活率。方法选用成年健康SD大鼠,于第3~4肋间开胸0.5~1cm,迅速暴露心脏,冠状动脉下穿线后,即刻复位关胸,提线时以标Ⅱ导联心电图ST段和T波的改变作为阻断血流评定指标,连续缺血15min,松线,缝合切口。结果改进后的造模成功率达到90%以上,高于传统方法的成功率(60%)。结论改进后的方法能大大降低手术难度,提高模型成功率和动物成活率,准确地复制心肌缺血再灌注损伤的实验模型,简化实验操作步骤。
Objective: To improve and evaluate the method of establishing rat models of acute myocardial ischemia reperfusion, which had high rate of success and survive. Methods: SD rats performed the left - sided thoracotomy form the third to fouth intercostal space to open the thoracic cavity 0.5 - 1.0 cm and exposed the heart. To pass through the cardiac muscle under the left coronary artery with a thread and returned the heart to the thoracic cavity then clamp the incisal opening immediately. The change of the electrocardiogram of lead I/could be regraded as the index of the blockage of blood flow when tensioned the thread. Slacked off the thread after kept the situation 15 min then sature the incisal opening. Results: The successful rate of the improved method was above 90% and its rate of traditional method was only 60%. Conclusion: The method of establish rat models of acute myocardial ischemia reperfusion is simple and convenient, with high successful rate and survival rate.
