人乙型肝炎病毒(ayw型)全基因组转基因小鼠病理学观察 被引量:4

摘要 目的:观察乙型肝炎病毒(HBV)ayw亚型全基因组转基因小鼠的肝、肾等组织的病理改变。方法:选取24只13~25周龄的清洁级ICR品系HBV转基因小鼠及15只正常ICR小鼠为对照进行病理大体观察,并重点取肝、肾、脾、肺、心、脑、睾丸(卵巢)、唾液腺、肌肉和皮肤等组织,切片HE染色后进行光镜和透射电镜形态观察。结果:上述两种清洁级小鼠大体和光镜下各器官和组织未见有明显病变;两种小鼠移入普通级环境下饲养4周后肝脏等器官发生机会性感染的机会相似;透射电镜下所有被检转基因小鼠的部分肝细胞扩张的滑面内质网等处有HBsAg样颗粒和Dane样颗粒,部分转基因小鼠之肾小管上皮细胞内见类似病毒颗粒;肝、肾细胞内未见其他亚微病理损害。结论:所建成的HBV转基因小鼠既能在肝(其次肾)细胞内产生HBV样病毒颗粒,但又不产生明显病理损害。 Objective:To make pathologic observation on HBV(ayw) intact genome transgenic mice(TGM) established by our cooperative research group.Methods:The TGM(24 cases,13-25 weeks) of ICR line in sterile grade and corresponding control mice (15 cases,13-25 weeks)were chosen for the study(some of them were transferred from sterile grade environment to common grade one for 4 weeks before pathologic examination).The liver,kidney,spleen,lung,heart,brain,testis /ovary,salivary glands,muscle and skin for each mouse were taken for gross pathologic examination,histopathologic examination under light microscope and ultrastructural pathologic examination under transmission electron microscope.Results:(1) No any pathologic changes were found by gross and histologic examination for TGM and control mice kept in sterile environment before sacrifice;(2) The occurrence rates of nonspecific inflammatory foci in livers were comparable for TGM and their controls when they were in common environment;(3)The HBsAg like particles,Dane like particles were found in the livers of all TGM and were distributed in the dilated smooth endoplasms and the cytoplasm of some hepatocytes without any other obvious pathologic changes;(4)These HBV like particles were also observed in some renal epithelia of several TGM cases.Conclusion:The TGM established herein can produce HBV like virus particles in the livers and some kidneys. But this did not result in other significant pathologic changes.
