摘要
[目的]探讨姬松茸在富硒过程中的生化机理。[方法]在姬松茸菌丝体培养基中加入浓度分别为03、0、609、01、201、50 mg/L的亚硒酸钠溶液,研究不同浓度硒对姬松茸菌丝体内过氧化物酶(POD)和超氧化物歧化酶(SOD)的活性以及丙二醛(MDA)含量的影响,[结果]当硒浓度为150 mg/L时,姬松茸菌丝体POD活性最高,为241.53μmol/(g.min)。当硒浓度为60 mg/L,姬松茸菌丝体SOD活性和MDA含量最高。低浓度硒能显著提高姬松茸菌丝体内POD和SOD活性、明显降低MDA含量,增强菌丝体的抗氧化能力。高浓度硒则对姬松茸菌丝体有毒害作用。[结论]在培养富硒姬松茸菌丝体过程中,应选取适宜的硒浓度。
[Objective] The research aimed to discuss the biochemical mechanism of Agaricus blazei Murill.in the process of enriching selenium.[Method] Sodium selenite solution with the concentration of 0,30,60,90,120,150 mg/L were added to the medium of A.blazei mycelia to study the effects of different concn.of selenium on the activities of peroxidase(POD) and superoxide dismutase(SOD),malondialdehyde(MDA) content in the mycelium of A.blazei.[Result] When the concn.of selenium was 150 mg/L,POD activity in the mycelium of A.blazei.was highest,being 241.53 μmol/(g·min).When the concn.of selenium was 60 mg/L,SOD activity and MDA content in the mycelium of A.blazei.was highest.Low concn.of selenium could increase the activities of POD and SOD in the mycelium of A.blazei.significantly,decrease MDA content obviously and strengthen the antioxidant capacity of mycelium.While high concn.of selenium had poisonous effects on the mycelium of A.blazei.[Conclusion] In the culture process of the mycelium of A.blazei,suitable concn. of selenium should be taken.
