摘要
目的:研究人类白血病细胞株及原代白血病细胞中人端粒结合蛋白2(TRF2)的表达。方法:应用实时定量RT-PCR,检测白血病细胞株及原代白血病细胞中TRF2 mRNA的表达,应用Western Blot,检测TRF2蛋白的表达。结果:T淋巴系白血病细胞株的TRF2表达显著高于骨髓单个核细胞和髓系白血病细胞株。在原代白血病细胞中,M0和M1亚型患者白血病细胞TRF2的表达高于其他各亚型急性髓系白血病(AML)患者。结论:预后差的AML患者和T细胞恶性肿瘤细胞株高表达TRF2,提示TRF2高表达可能和白血病的预后有关。
Objective: To detect the expression levels of telomere binding factor 2 (TRF2) on leukemia cell lines and primary leukemia cells. Methods: The expression of TRF2 mRNA was detected with quantitive real-time RT-PCR in leukemia cell lines and primary leukemia cells. The Western blot analysis was used for the detection of TRF2 protein expression. Results: TRF2 was overexpressed in T-cell leukemia cell lines but not in myelogenous leukemia cell lines. Significant higher expression levels of TRF2 were observed in primary leukemia cells from patients with M0 and M1 subtypes of acute myelogenous leukemia (AML) compared with normal control and other subtypes of AML. Conclusion. Increased TRF2 expression levels are found in T-cell leukemia cell lines and AML patients with poor prognosis,which suggests that TRF2 expression might be related to the prognosis of leukemia.
