摘要
目的探讨血管紧张素Ⅱ(AngⅡ)和胰岛素对人脐动脉平滑肌细胞产生一氧化氮(NO)、超氧阴离子(O2-)、三磷酸肌醇(IP3)活性的影响。方法不同浓度的AngⅡ和胰岛素加入体外培养的人脐动脉平滑肌细胞,分别测定细胞培养液中一氧化氮合酶(NOS)、超氧化物歧化酶(SOD)活性,NO、环鸟苷酸(cGMP)和O2-浓度,3H标记肌醇-逐步洗脱法测定IP3活性。结果胰岛素(10、100、1000mU/L)使平滑肌细胞NOS活性升高,NO、cGMP产生增加,对SOD活性和O2-水平无影响,对IP3活性无明显影响;0.1、0.5nmol/LAngⅡ对NOS活性、NO和cGMP产生无明显影响,1nmol/LAngⅡ使NOS活性下降(P<0.05),NO、cGMP产生减少(P<0.05或<0.01),0.1、0.5、1nmol/LAngⅡ使SOD活性下降(P<0.05或<0.01),O2-水平升高(P<0.01),使IP3活性升高(P<0.05);胰岛素10mU/L+AngⅡ0.1nmol/L、胰岛素100mU/L+AngⅡ0.5nmol/L、胰岛素1000mU/L+AngⅡ1nmol/L使NOS活性下降(P<0.01),NO和cGMP产生减少(P<0.01),SOD活性下降(P<0.01),O2-水平升高(P<0.01),使IP3活性升高(P<0.05);0.1nmol/LAngⅡ+1μmol/L氯沙坦、0.5nmol/LAngⅡ+5μmol/L氯沙坦、1nmol/LAngⅡ+10μmol/L氯沙坦可以改善AngⅡ引起的NOS、SOD活性下降,NO、cGMP产生减少,O2-水平升高,以及AngⅡ引起的IP3活性升高,但与对照组比较,差异无统计学意义(P>0.05)。结论胰岛素使平滑肌细胞NO产生增加,对IP3活性无影响。高浓度AngⅡ使NO产生减少,O2-水平升高,使IP3,活性升高,可以被氯沙坦阻断,胰岛素和AngⅡ同时存在时平滑肌细胞NO产生减少,O2-水平升高,可以使IP3活性升高。
Objective To investigate the effects of angiotensin Ⅱ and insulin on nitric oxide (NO), superoxide anion and inositol trisphosphate (IP3) activities secreted by human umbilical artery smooth muscle cells (HUASMCs). Methods HUASMCs were incubated with angiotensin Ⅱ and insulin respectively for 24 hours. In the end of the experiment, NOS and SOD activity, the concentration of NO, cGMP and O2^- in HUASMCs supematant was measured, IP3 activity was determined by 3H-labeled inositol with step by step elution method. ResUlts NOS activity,the concentration of NO and cGMP increased in the presence of 10, 100, 1 000 mU/L insulin, while SOD activity and the concentration of O2^-didn' t change significantly; IP3 activity in SMC didn't change significantly. 0.1,0.5 nmol/L Ang Ⅱ had no significant effect on NOS activity and the concentration of NO, cGMP, while NOS activity and the concentration of NO, cGMP decreased in the p iesence of 1 nmol/L Ang Ⅱ;SOD activity decreased while the concentration of O2^- increased in the presence of 0.1, 0.5, 1 nmol/L Ang Ⅱ ; IP3 activity increased in the presence of 0.1,0.5 nmol/L, 1 nmol/L AngⅡ ; NOS, SOD, activity, the concentration of NO, cGMP decreased, IP3 activity increased while O2^-increased when SMC treated with in sulin 10mU/L + Ang Ⅱ 0.1 nmol/L, insulin 100mU/L+ Ang Ⅱ 0.5 nmol/L,insulin 1 000 mU/L + Ang Ⅱ 1 nmol/L; losartan could reverse the effects of Ang Ⅱ . Conclusion Insulin could increase NO concentration in HUASMCs, but which didn't decrease IP3 activity, while O2^-increase and IP3 activity increased in the presence of AngⅡ , which could be inhibited by lorsartan, while O2^- increased more significantly, IP3 activity increased when HUASMC is treated with both Ang Ⅱand insulin,
