摘要
目的:通过检测商陆粗提物对大鼠阿霉素肾病模型大鼠的血、尿蛋白、肾组织一氧化氮(NO)、血清可溶性白细胞介素2受体(sIL-2R)的影响。观察商陆对阿霉素肾病大鼠的治疗作用,为开发中药商陆提供依据。方法:经大鼠尾静脉分次注射阿霉素合计7.5mg/kg,制备肾病模型,分别灌以商陆粗提物4g·kg-1·d-1和1g·kg-1·d-1以及泼尼松16mg·kg-1·d-1,连续2周,观察用药组间动物血清白蛋白、尿蛋白、sIL-2R及肾组织NO的变化。结果:(1)商陆粗提物对阿霉素肾病模型大鼠具有减少尿蛋白、提高血清白蛋白的作用(与模型组比较,P<0.05),且与激素比较无统计学差异(P>0.05);(2)sIL-2R水平各治疗组均下降,其中高剂量商陆组与模型组差异明显(P<0.05);(3)商陆各治疗组肾组织内NO浓度降低(与模型组比较,P<0.05),且与激素比较无统计学差异(P>0.05)。结论:(1)商陆可以减少阿霉素肾病大鼠尿蛋白排出,提高阿霉素肾病大鼠血清白蛋白的水平。(2)商陆各治疗组在尿蛋白排出减少、血清白蛋白的水平提高的同时血清sIL-2R水平降低、肾组织内NO浓度降低,提示其减少尿蛋白排出、提高血清白蛋白的水平作用可能与商陆可以下调血清sIL-2R水平或降低肾组织内NO水平有关。
Objective: To observe the effects of crude extract from phytolaecae on adriamiein - induced nephropathy rats and the change of serum - albumin, Urine protein ,nitrogen monoxidum(NO) ,slL- 2R of the rats. To observe the therapeutical effect of phytolaccae on adriamiein - induced nephropathy rats , To provide experimental data for clinical use of phytolaccae to treat such patients. Methods:Male SD rat nephropathy was induced by multiple injection of adriamiein to vena caudalis, with total dose 7.5 mg/kg. Then the model rats were fed with crude extract from phytolaccae(4 g· kg^-1 d^-1 or 1 g· kg^-1 d^-1 ) or prednisone ( 16 mg· kg^-1 d^-1 ) for 2 weeks. Serum albumin, urine protein , nitrogen monoxidum and slL- 2R were measured. Results: ( 1 )The crude extract from phytolaccae can decrease urine protein, devate serum albumin(compared with rnodel group P〈0.05). There is no statistic difference between the effects of the extract and prednisone ( P 〉 0.05 ). (2) The slL - 2R decreased in all treated groups, with a significant decrease in high dosage group ( P 〈 0.05 compared with model group). (3) The kidney tissue NO were decreased in both phytolaccae and prednisone groups ( P 〈 0.05 compared with model group), and there was no statistical difference between the two groups ( P 〉 0.05). Conclusion: (1) The crude extract from phytolaccae can decrease urine protein, devate serum albumin. (2)With the decrease of urine protein, the boost of serum albumin, The slL- 2R decreased and The kidney tissue NO decreased were discovered. There are possibility that the decrease of urine protein and the boost of serum albumin correlate to that the crude extract from phytolaccae can decrease the slL- 2R in the blood serum or the NO in kidney tissue.
