人工感染仔猪猪生殖与呼吸综合征病毒的核酸分布规律 被引量:4

摘要 根据猪生殖与呼吸综合征病毒(PRRsV)美洲株ATCCVR-2332保守区ORF6和ORF7的基因序列,设计了1条37bp的寡核苷酸探针,末端用生物素标记。应用原位杂交方法对人工感染PRRSVSC-1株的28日龄仔猪进行了病毒核酸分布规律的研究。结果显示,感染后7~28d于肝、脾、肺、肾、扁桃体、胸腺、肺门淋巴结、肠和大脑检测到阳性杂交信号,其中肺门淋巴结的阳性信号一直较强,脾的阳性信号逐渐增强,肺的阳性信号较弱,心脏未见阳性杂交信号。 According to the conserve ORF6 and ORF7 gene sequences of the North American strain ATCC VR-2332 of porcine reproductive and respiratory syndrome virus(PRRSV),a 37 bp oligonucleotide probe was designed,and labeled with biotin. The distributions of PRRSV in the 28-day-old-piglets at different time points post-infection were analyzed by using in situ hybridization. The result showed that positive signals were found from day 7 to 28 post-infection in tonsil, hilar node, thymus, spleen, lung, kidney, and cerebrum. Strong positive in situ hybridization signal was found in hilar node. The positive signals in spleen increased gradually post-infection. The weak positive signals were found in lung. Signal in heart was negative.
