摘要
目的探讨兔骨髓间充质干细胞(BMSCs)在体外诱导成软骨细胞的生物学特性,为骨组织工程提供实验依据。方法体外培养获取生长状态良好的兔骨髓间充质干细胞,进行成软骨诱导,观察并检测其生物学特性。结果BMSCs在转化生长因子-β1、维生素C的作用下,7d后可见细胞多变为圆形或椭圆形,并聚集形成软骨样结节,阿利辛蓝染色、蕃红花"O"-亮绿染色显示细胞内酸性粘多糖含量高,Ⅱ型胶原免疫组织化学染色阳性,PCR检测有Ⅱ型胶原的表达,表现出软骨细胞的分化特点。结论在特定诱导条件下,兔BMSCs体外可定向诱导分化为软骨细胞,为骨组织工程提供实验依据。
Objective To study the bone tissue chondrogenic ability of rabbit bone marrow stem cells(BMSCs) in vitro in order to provide the experimental evidence for tissue engineering. Methods Rabbit BMSCs were cultured in vitro induced to chondrocytes,and then the biologic characters of the chondrocyte were observed and tested. Results BMSCs became round or ovoid from spindle shape after 7 days exposure to TGF-β1 and vitamin C in conditional culture, then turned into chondrogenic nodules. The test of Alcian blue staining, saffron "O" and immunohistochemistry staining for type Ⅱ collagen show positive. The test of PCR shows the expression of ,Ⅱ collagen, all revealed the characteristics of chondrogenic differentiation. Conclusions Rabbit BMSCs may be induced into chondrocytes under conditional culture media in vitro.
