摘要
通过分光光度法对池蝶蚌的抗菌力,溶菌酶和酚氧化酶活性进行了分析。结果表明,经嗜水气单胞菌感染后,池蝶蚌对这两种细菌的抗菌力分别为0.61±0.01 U和0.14±0.02U;所有实验组对大肠杆菌和金黄色葡萄球菌均无抑制效果。随着年龄的增长,溶菌酶活性也显著增加,第4龄蚌的溶菌酶活力可达0.93±0.05U,比1龄蚌的酶活力高一倍左右;十二烷基磺酸钠(SDS)能够激活酚氧化酶(PO)酶原系统,2龄前,酚氧化酶活力逐渐增强,3龄和4龄逐渐减弱,2龄蚌的PO活性最高,为22.70±4.39 U。研究表明,池蝶蚌血淋巴中免疫因子的种类和表达活性在个体不同发育阶段中呈现出差异,推测可能与水体环境对蚌的作用有关。
Hyriopsis schlegeli was high quality freshwater mussel from Japan. For the sake of knowing the characteristics of anti - disease, the mechanism of freshwater mussel innate immunity and selecting disease - resistance variety. It was necessary to investigate Hyriopsis schlegeli immunity. The activities of the antibacterial, bacteriolysis and phenoloxidase were considered as significant index in shell immunity. They were investigated with the methods of spectrophotometer. The antibacterial activities to Aeromonas hydrophila and Bacillus subtilis in the haemolymph of healthy Hyriopsis schlegeli were 0.83 ± 0.01 U and 0.34 ± 0.01 U respectively, which were superior to that of healthy Hyriopsis cumingii ( 0. 57 ± 0. 03 U and 0.26 ± 0.01 U) and the injected bacterial group ( 0.61 ± 0.01 U and 0.14 ± 0.02U ). The antibacterial activities to Escherichia coli and Staphylococcus aureus were not detected among three groups. The experimental data of lysozyme activities indicated that the activities increased with age, the activity of 4 - year mussel was the highest ( 0.93 ± 0.05 U). The Pro - phenoloxidase system was stimulated by SDS, and the phenoloxidase activity was increased before 2 - year - old while decreased gradually later. The phenol- oxidase activity of 2 - year was in the peak among 4 years mussel. The pattern and activities of immunity factors varied with development stage, it maybe related to the effect of environmental factor in water.
