摘要
目的:探讨不同染氟剂量在不同时间段对成纤维细胞和成骨细胞c-fos和c-jun基因表达的影响.方法:应用RT-PCR和琼脂糖凝胶电泳方法检测和比较染氟对成纤维细胞和成骨细胞c-fos和c-jun基因表达的影响.结果:染氟可促进成纤维细胞c-fos基因表达,在过量氟的刺激下,成纤维细胞和成骨细胞c-fos基因表达均有增强.染氟对成纤维细胞c-jun基因表达增强作用不明显.结论:染氟成纤维细胞已有成骨表型表现,在氟刺激成纤维细胞向成骨细胞转化过程中,c-fos也发挥重要作用.
Objective: To study the effects of fluoride on the gene expression of c- los and c- jun in fibroblast and osteoblast at the different dosages and intervals. Methods: The effects of fluoride on the gene expression of c - los and c - jun in fibroblast and osteoblast were detected and compared with RT - PCR and agarose gel electrophoresis. Results: Upgraded gene expression of c - los in fibroblast exposed to fluoride was shown. Gene expression of c- los in both fibroblast and osteoblast exposed to overdosage of fluoride was heightened. No manifest enhancing effect of fluoride on the gene expression of c- jun in fibroblast. Conclusion: Fibroblast when exposed to fluoride shows the phenotypic expression of osteoblast. Gene c - los plays an important role in the transformation of fibroblast to osteoblast.
