摘要
目的确定一株蛇足石杉内生真菌2F09P03B发酵培养物中石杉碱甲的超声波辅助提取工艺,以改善石杉碱甲提取率。方法对提取溶剂、pH值等影响提取效率的因素分别进行单因素实验;对料液比、超声波功率、负载率(dutycycle)、超声时间4个影响超声波提取效率的因素在单因素实验基础上利用正交实验进行优化。结果超声波辅助提取石杉碱甲的最佳工艺条件是:提取溶剂为pH4.0的酸水,碱性还原反应在pH7.5~8.0进行,料液比1:20,超声波功率140W,超声波负载率30%,超声时间40min。此条件下,该内生真菌石杉碱甲的提取率达0.074%,约为其他传统提取方法的1.5~2倍。结论超声波辅助提取法的石杉碱甲提取率较其他方法有了显著提高,并能大幅度缩短提取时间,应在蛇足石杉内生真菌2F09P03B石杉碱甲的提取中加以应用。
OBJECTIVE To investigate the ultrasonic extraction condition of huperzine A from an endophytic fungus 2F09P03B isolated from Huperzia serrata which can pruduce the same active component as its parasitifer does, with the aim of elevating the extraction ratio of huperzine A. METHODS The factors that affected the extraction of huperzine A, including extraction solvents, methods and pH were investigated respectively by single factor method. Other factors that affected the ultrasonic extraction, including ratio of material to liquid, ultrasonic power, duty cycle, ultrasonic time, were optimized by orthogonal experiment. RESULTS The suitable extraction solvent was at pH 4. 0. The optimal condition of reduction reaction was pH 7.5 - 8.0. The optimum ultrasonic extraction factors were as follows the ratio of material to liquid was 1: 20, ultrasonic power was 140 W, ultrasonic duty cycle was 30%, ultrasonic extraction time was 40 min. Under this condition, the average extraction ratios of huperzine A was 0. 074% , as 1.5 -2. 0 times as that of other methods. CONCLUSION Compared with traditional extraction methods, the ultrasonic extraction method can elevate the extraction ratio of huperzine A and reduce the process time remarkably, worth being applied to the extraction of huperzine A from endophytic fungus 2F09P03B.
