摘要
应用基因工程技术,对本室已经克隆的日本血吸虫成虫32kDa蛋白分子的cDNA片段进行PCR扩增,扩增产物亚克隆入高效真核表达载体pCD,构建成功裸露DNA疫苗pCDSj32。pCDSj32在BALB/c小鼠骨骼肌细胞得到表达。表达产物可被小鼠抗32kDa蛋白分子单抗识别。免疫荧光定位显示,表达产物不但存在于细胞浆,而且可结合于胞膜并被分泌至胞外。
The specific cDNA segment of 32kDa protein in Schistosoma japonicum was amplified with PCR. The product was subcloned into a high level eukaryotic expression vector pCD, then, naked DNA vaccine pCDSj32 was constructed. pCDSj32 was expressed in the skeletal muscular cells of BALB/c mice. The expressed products could be specially recognized by sera from rabbits infected with Schistosoma japonicum and monoclonal antibody to 32kDa protein. The expressed protein was not only located within the plasma of the cells, but also combined with the membranes and secreted out of the cells.
