半夏中抑制肿瘤坏死因子-α产生的新脑苷 被引量:19

摘要 目的研究半夏Pinellia ternata鲜块茎中的生物活性成分,为其质量评价提供科学依据。方法采用多种柱色谱方法分离纯化,根据理化性质和谱学方法鉴定结构。采用MTT法,体外观察化合物对脂多糖(LPS)刺激的小鼠腹腔巨噬细胞产生肿瘤坏死因子-α(TNF-α)的抑制作用。结果分离得到8个化合物,分别鉴定为环阿屯醇(I)、β-谷甾醇(Ⅱ)、α-棕榈精(Ⅲ)、胡萝卜苷(Ⅳ)、异半夏苷(1-O-β-D-吡喃葡糖基-(2S,3R,4E,11Z)-2-[(2R-羟基十六碳烯酰基)氨基]-4,11-十八碳二烯-1,3-二醇,V)、胡萝卜苷-6′-O-二十烷酸酯(β-谷甾醇-3-O-β-D-葡萄糖苷- 6′-O-二十烷酸酯,Ⅵ)、尿嘧啶(Ⅶ)和蔗糖(Ⅷ)。环阿屯醇和异半夏苷在10μmol/L对LPS刺激的小鼠腹腔巨噬细胞产生TNF-α的抑制率分别为54.7%和47.5%。结论异半夏苷为新化合物,半夏抗炎作用可能与环阿屯醇和异半夏苷有关。 Objective To study the chemical constituents in the fresh rhizome of Pinellia ternata, and provide the scientific basis for quality control. Methods Various chromatographic techniques were used to separate and purify the chemical constituents and their chemical structures were determined on the basis of physicochemical properties and spectroscopic analysis. The inhibitory effects of the isolated compounds on tumor necrosis factor-α (TNF-α) production in the peritoneal macrophages of mice stimulated with lipopolysaccharide (LPS) were observed in vitro by microplate ultraviolet-colorimetric method. Results Eight compounds were isolated and identified as cycloartenol (Ⅰ), β-sitosterol (Ⅱ), α-monpalmitin (Ⅲ), daucosterol (Ⅳ), isopinelloside (1-O-β-D-glucopyranosyl-(2S, 3R, 4E, 11Z)-2-(2R-hydroxyhexadecenoyl-amino)-4, 11-octadecadiene-1, 3-diol, Ⅴ), daucosterol-6'-O-eicosanate (β-sitosterol-3-O-β-D-glucoside-6'-O-eicosanate, Ⅵ), uracil (Ⅶ), and sucrose (Ⅷ). The inhibitory effects of cycloartenol and isopinelloside were 54.7% and 47.5%, respectively, against the TNF-α production in the peritoneal macrophages of mice stimulated with LPS at a concentration of 10 μmol/L in vitro. Conclusion The isopinelloside is a new compound. The anti-inflammatory effects of the rhizome of P. ternata is at least related to cycloartenol and isopinelloside.
